Stoke’s initial focus for its TANGO platform is on diseases of the central nervous system and the eye that are caused by a loss of ~50% of normal protein levels (haploinsufficiency).

Stoke has identified STK-002 as a clinical candidate for the potential treatment of autosomal dominant optic atrophy (ADOA), the most common inherited optic nerve disorder. The company is also pursuing a potential new medicine for Syngap1, a severe and rare neurodevelopmental disorder, in collaboration with Acadia Pharmaceuticals.

Proof of concept has been demonstrated in other organs, tissues, and systems, supporting broad potential for the Company’s proprietary approach.

                              

Position Purpose: 

We are seeking experienced Associate Scientist who will support ongoing research efforts in designing and evaluating antisense therapies for a diverse group of CNS diseases. This position will report to a Principal Scientist in the in vitro pharmacology group. As part of the group you will work collaboratively to design and perform gene and protein expression assays to support development of new ASO-based therapies.

Key Responsibilities:

  • Work in aseptic conditions and maintain cultures of various cell types. Experience with iPSC-derived CNS cell types is a plus.
  • Evaluate oligonucleotide-based drug candidates in cell-based assays.
  • Conduct in vitro disease model cellular assays to evaluate drug candidates.
  • Design, execute and troubleshoot routine experiments independently.
  • Perform cell and molecular biology techniques such as RNA and protein isolation, RT-PCR, qPCR, western blotting and other project-specific assays, as necessary. Familiarity with ELISA and capillary-based protein assays in a plus.
  • Use microscopy and immunohistochemistry-based methods to evaluate cellular phenotypes.
  • Maintain accurate documentation of experiments in electronic lab notebook (ELN).
  • Analyze and interpret data independently.
  • Author reports and contribute to manuscripts.
  • Present research findings at group meetings.
  • Work independently as well as collaborate with peers in a result-oriented research environment.

Required Skills & Experience: 

  • S with a minimum of 7 years post-graduate experience or M.S. with a minimum of 4 years of experience in neuroscience, biotechnology, cell and molecular biology or related field, preferably in the biotech or pharma industry.
  • Strong background in cell and molecular biology research; understanding of the basics of PCR is a must.
  • Strong Hands-on experience in RNA isolation techniques, mammalian cell culture and cell-based assays is strongly desirable.
  • Strong organizational and recordkeeping skills is a must. 
  • Ability to adapt to changing priorities and deadlines.
  • Excellent planning and time management skills including the ability to support and prioritize multiple projects.

This position is an on-site position located in Bedford, MA location.
