Stoke’s initial focus for its TANGO platform is on diseases of the central nervous system and the eye that are caused by a loss of ~50% of normal protein levels (haploinsufficiency).
Stoke has identified STK-002 as a clinical candidate for the potential treatment of autosomal dominant optic atrophy (ADOA), the most common inherited optic nerve disorder. The company is also pursuing a potential new medicine for Syngap1, a severe and rare neurodevelopmental disorder, in collaboration with Acadia Pharmaceuticals.
Proof of concept has been demonstrated in other organs, tissues, and systems, supporting broad potential for the Company’s proprietary approach.
Position Purpose:
We are seeking an enthusiastic and experienced bioinformatician to join the computational group focusing on mRNA splicing and antisense oligonucleotides. Drug discovery at Stoke begins with computational biology. As part of the early discovery team led by Stoke’s co-founder, you will work collaboratively with data and experimental scientists to expand our analytical approaches in identifying therapeutic targets relevant to the TANGO technology.
Key Responsibilities:
- Assist in the development of strategic plans for analyzing and deciphering next-generation sequencing data, and apply modern computational biology approaches to advance Stoke's portfolio of therapeutic targets.
- Propose and develop analytical and visualization tools to deliver complex biological data.
- Build, deploy, and maintain bioinformatics applications on Amazon Web Services.
- Contribute to the development and maintenance of web applications to facilitate target identification and data analysis.
- Use computational tools to investigate novel approaches to upregulate gene expression.
- Provide program-specific support for pre-clinical and clinical groups, such as biomarker development and mechanism of action of ASO clinical candidates.
- Support a highly collaborative environment and promote positive teamwork among a team of scientists from different backgrounds.
- Communicate scientifically rigorous results clearly, verbally and in writing, both internally and externally.
Required Skills & Experience:
- A degree in bioinformatics, computational biology, computer science, mathematics, systems biology, or any related quantitative field.
- Previous experience in the biotech or pharma industry; 14 years with a bachelor’s degree, 8 years with a master’s degree, or 4 years with a Ph.D.
- Experience in and deep understanding of posttranscriptional gene expression regulation, including but not limited to RNA splicing, RNA surveillance. pathways, RNA binding proteins, and antisense oligonucleotides.
- Fluency in coding languages such as Python and R, including scientific modules and packages commonly used in bioinformatics. Familiarity with machine learning algorithms and applications is a plus.
- Direct experience in developing and maintaining reproducible analytical workflows for high-throughput sequencing assays such as bulk or single-cell RNA-Seq, eCLIP, or whole-genome sequencing using workflow management systems such as Snakemake or Nextflow.
- Management experience in leading computational scientists and interns.
Location(s):
This position is a hybrid position with an office setting based in our Bedford, MA location.
Travel:
This position will require approximately 5% travel.
